How does Sinetrol® support fat metabolism?
One major factor in healthy weight management is the body’s ability to release stored fat. This process is called lipolysis, and it involves the breaking down of triglycerides stored inside the fat cells into free fatty acids and glycerol, allowing those fatty acids to be released and used as a fuel source for our bodily processes.
One proposed mechanism involves the positive impact it has on cAMP, an important signalling molecule involved in telling fat cells to release some of their stored energy to be transformed into ATP (our body’s energy currency) by the mitochondria. Phosphodiesterase enzymes (such as PDE3B) normally shorten this signal by breaking cAMP down. Laboratory and animal research suggests that Sinetrol® can inhibit PDE3B-related activity, supporting the cAMP signal to remain active for longer. This is associated with increased activity of proteins involved in fat mobilisation, including adipose triglyceride lipase (ATGL) and perilipin (PLIN).2,3
In human adipocytes, Sinetrol® increased free-fatty-acid release approximately six-fold compared with control and showed around 2.1 times greater lipolytic activity than guarana alone, despite providing less caffeine.2
In simple terms, Sinetrol® may help the body become better at mobilising stored fat so it can be made available for energy.
What does the clinical research show?
Several human trials have investigated Sinetrol® in overweight individuals.
In an early placebo-controlled study, 12 weeks of supplementation was associated with a 15.6% reduction in body fat and an average 5.2kg reduction in body weight.2 Although this trial was small, later studies have continued to show benefits for body composition.
A larger 12-week randomised, double-blind, placebo-controlled trial involving 95 adults found greater reductions with Sinetrol® than placebo in:4
- Body fat – approximately 9.7%
- Body weight – 2.62kg
- Waist circumference – approximately 5.2cm
- Hip circumference – approximately 5.2cm
Circulating free fatty acids also increased by around 330%, supporting the lipolytic effect. The study additionally reported reductions in circulating blood glucose levels, CRP (an inflammation associated marker), fibrinogen (a protein involved in blood clotting and inflammation), and malondialdehyde (a highly reactive end-product of lipid oxidation), and improvements in major antioxidant markers (increased superoxide dismutase and glutathione levels).4
Why abdominal fat matters
Visceral fat surrounds the abdominal organs and behaves as an active endocrine tissue. Higher levels are strongly associated with metabolic dysfunction and inflammatory signalling, making waist circumference and abdominal fat particularly useful markers during weight management.
In a 12-week pilot trial in overweight men, supplementation with 630mg Sinetrol® was associated with a 9.7% reduction in abdominal fat, alongside reductions of approximately 7.4cm from the waist and 5.6cm from the hips.5
A longer 16-week study also reported reductions in body fat, with around 65% of total fat loss coming from the trunk area.6
Can Sinetrol® help preserve muscle?
This is probably one of the most important factors to consider because losing weight doesn’t automatically mean improving body composition. If significant muscle tissue is lost alongside fat, strength and metabolic resilience may also decline. More recent research also found an improved lean-to-fat mass ratio without measurable lean-mass loss, alongside a 5.2% reduction in body fat.6
